🧬 A complementary approach for advanced solid tumors The greatest potential for WEE1 and PKMYT1 inhibition lies in cancers marked by high replication stress and genomic instability, where late cell-cycle checkpoints become critical for survival. This includes advanced solid tumors with specific molecular vulnerabilities, such as alterations that drive uncontrolled cell-cycle progression and checkpoint dependence. Tumor types where these features are commonly observed include: ovarian, breast, lung, colorectal, gastric, and pancreatic cancers, among others. By aligning therapy with underlying biology, this combination strategy aims to focus treatment where it is most likely to be effective. 👇 Learn more about our ongoing clinical trial https://lnkd.in/dfAtKP2M #DNADamageResponse #DDR #PKMYT1 #WEE1 #Oncology #ClinicalTrial #Debiopharm

🧬Zedoresertib and lunresertib: a unique double hit outcome Cancer cells depend on cell-cycle checkpoints to survive replication stress. The combination of WEE1 and PKMYT1 inhibition completely topples the crucial G2/M checkpoint where cells would normally pause to repair DNA damage and replication stress. With both checkpoints disabled, a lethal domino effect begins: unrepaired damage is carried into division, cascading into mitotic catastrophe and cell death. Zedoresertib and lunresertib provide complementary mechanisms of action, representing a novel synthetically lethal approach with strong preclinical synergy. 👇 Read more about Debiopharm’s work on this unique combination https://lnkd.in/dvY6FwyV #DNADamageResponse #DDR #PKMYT1 #WEE1 #Oncology #Debiopharm

🧬The MYTHIC trial This Phase 1/1b, multi-center, open-label, dose-escalation study is currently being executed in biomarker-selected patients (CCNE1 amplification or deleterious FBXW7/PPP2R1A alterations), reflecting a shared biology of heightened replication stress and checkpoint dependency. (NCT04855656) Module 4 of the study is designed to evaluate safety and tolerability, define MTD and RP2D, characterize PK and pharmacodynamics, and assess preliminary anti-tumor activity for lunresertib in combination with zedoresertib. As presented at AACR 2026, early data readouts from MYTHIC suggest synthetic lethality between lunresertib and zedoresertib, with consistent tumor regressions observed. Strong signals of activity were observed with the combination, particularly in patients with platinum resistant or refractory ovarian cancer. 👇 Read more about lunresertib and the MYTHIC trial https://lnkd.in/dUeS9ztr https://lnkd.in/dfAtKP2M #DNADamageResponse #DDR #PKMYT1 #WEE1 #Oncology #ClinicalTrial #Debiopharm

🧬Introducing lunresertib In July 2025, Debiopharm acquired the exclusive worldwide rights to lunresertib from @Repare Therapeutics. Lunresertib is a first-in-class, selective and potent, oral small-molecule inhibitor of PKMYT1, a cancer target identified as synthetically lethal in solid tumors with CCNE1 amplification and alterations in FBXW7 and PPP2R1A. Together with zedoresertib, lunresertib adds an additional row to Debiopharm’s DDR domino cascade. As a leading selective PKMYT1 inhibitor in clinical development, lunresertib has demonstrated encouraging activity across multiple clinical trials in difficult-to-treat solid tumors. Positive initial Phase 1 data from the ongoing MYTHIC trial (NCT04855656) provide proof of concept for lunresertib as monotherapy and in combination. 👇 Find out more about lunresertib https://lnkd.in/dUeS9ztr #DNADamageResponse #DDR #PKMYT1 #Oncology #Debiopharm

🧬 Targeting a single DDR dependency can be highly effective. WEE1 inhibition has demonstrated robust anti-tumor activity by exploiting the inherent vulnerabilities of cancer cells. But DDR signaling is not governed by a single pathway. Cancer cells rely on parallel and compensatory checkpoints to maintain survival under replicative stress. Combination strategies offer the potential to sharpen tumor selectivity and allow for a more balanced pathway inhibition. At Debiopharm, the next phase of our DDR story explores what combination therapies could unlock. 👉 Watch this space to learn more! #DNADamageResponse #DDR #CombinationTherapy #Oncology #Debiopharm
