Dilated cardiomyopathy is a leading cause of heart failure and heart transplantation. 🫀 BAG3 variants account for up to 5% of genetically resolved DCM, and the genetics are increasingly well understood. So why is it still so hard to find therapies? In large part, because the models haven't kept up. 2D cultures don't generate contractile force, and animal models don't fully reproduce human cardiac disease. For DCM, a disease of failing heart muscle, force is the phenotype that matters. Curi Bio and Greenstone Biosciences set out to measure it. We started with a patient-derived iPSC line carrying a BAG3 frameshift mutation, from a donor whose heart was pumping at a 12% ejection fraction. We built it into 3D human engineered heart tissues and profiled them on our Mantarray™ and Nautilai™ platforms. The tissues reproduced the patient's disease. Our study also raised the questions that matter most for therapy development: ➡️ Is the deficit in the heart's contractile machinery, or in how that machinery responds to calcium? ➡️ How does a diseased heart tolerate a drug that healthy models say is safe? ➡️ What can a single cohort of tissues reveal across multiple drug challenges and 28 days in culture? Join us on October 28, when Dr. Todd Herron and Dr. Thomas Leahy, PhD will share the answers, and what they mean for modeling inherited cardiomyopathies in human tissue. 📅 Wednesday, October 28 · 8 AM PDT (11 AM EDT / 4 PM CET) 🔗 Register today: https://hubs.ly/Q04z5ddF0 #DilatedCardiomyopathy #HeartFailure #CardiacSafety #iPSC #3DTissueModels #RareDisease #NAMs #DrugDiscovery #CuriBio #GreenstoneBiosciences

What does 3D contractility reveal that 2D models can miss? Join Curi Bio and Greenstone Biosciences on October 28 for new data on genotype-anchored cardiac disease modeling using BAG3-mutant human iPSC-derived cardiomyocytes. Using 3D engineered heart tissues profiled on Mantarray™, we observed a dilated cardiomyopathy-like functional phenotype: ✔️ Lower contractility ✔️ Loss of pacing fidelity ✔️ Higher calcium needed to generate force, and ✔️ Reduced contractile reserve under verapamil If you’re building human-relevant cardiac models for your cardiac program, this is a webinar worth your time. 📅 Wednesday, October 28 🕗 8 AM PDT (11 AM EDT / 4 PM CET) 👉 Register today: https://hubs.ly/Q04yfdfj0 #DilatedCardiomyopathy #CardiacSafety #iPSC #3DTissueModels #NAMs #DrugDiscovery #CuriBio #GreenstoneBiosciences
