RNA editing meets DNA repair: new insights uncovered using AlidaBio’s EpiPlex™ platform New research published in Frontiers in Genetics reveals how ADAR1-mediated A-to-I RNA editing safeguards genome stability by regulating transcripts involved in DNA repair. Using Alida Biosciences’ EpiPlex platform, Yoshida et al. (千葉大学 Chiba University) profiled global A-to-I RNA editing in human TK6 cells and identified 870 edited genes, including 27 linked to DNA repair. Notably, bulk RNA-seq was not sensitive enough to resolve expression differences in these DNA repair genes. Key findings: → A-to-I RNA editing occurs in 3’-UTRs or introns of major DNA repair genes such as ATM, BRCA1, and XPA. → Loss of ADAR1 revealed gene- and locus-specific effects (altered XPA splicing, and reduced ATM mRNA stability). → ADAR1 plays a crucial role in genotoxic stress response (cells lacking ADAR1 showed increased sensitivity to cisplatin). This work highlights the interplay between RNA editing and genome maintenance, showing how epitranscriptomic regulation influences DNA repair, alternative splicing, and cellular stress resilience. Exciting insights from the Esteller and Macip labs were presented at the 2nd International Senescel Meeting in Barcelona. Their research shows how ibrutinib reshapes the aging brain by altering both transcriptomic and epitranscriptomic landscapes. Using the EpiPlex™ platform for epitranscriptomics, the study found that m⁶A and inosine RNA modifications change with aging and treatment, correlating with transcriptomic and proteomic shifts. These marks are enriched in specific biological clusters depending on the modification type, supporting their potential as a therapeutic strategy to ameliorate brain aging. Congratulations to Júlia Rius Bonet, Eloy Santos Pujol, Dr. Manel Esteller, and Dr. Salvador Macip for advancing our understanding of how epitranscriptomic regulation impacts neurobiology and aging. #Epitranscriptomics #RNA #AgingResearch #Neuroscience #m6A #Inosine #PrecisionBiology #EpiPlex

Profiling RNA Modifications in Scarce Clinical Samples: Mondal Lab Showcases EpiPlex at EMBO 2025 We’re proud to highlight new work from the Mondal Lab (University of Gothenburg), presented this week at the EMBO Workshop: The Epitranscriptome (28–30 October 2025). The team is investigating how RNA modifications influence cancer biology, focusing on epitranscriptomic regulation in high-risk neuroblastoma. Using their embryonic stem-cell–derived neuroblastoma model, they’re probing the functional role of m6A in tumor initiation and progression. To meet the challenge of extremely limited clinical material, the Mondal Lab turned to the EpiPlex platform, AlidaBio’s multi-modification profiling method optimized for low-input RNA. With EpiPlex, they can sensitively detect m6A and inosine across scarce tumor samples, capturing a more complete view of the epitranscriptomic landscape where traditional methods fall short. Congratulations to the Mondal Lab for advancing our understanding of how RNA modifications drive cancer biology, and for pushing the frontier of epitranscriptomic discovery. #Epitranscriptomics #Neuroblastoma #RNAmodifications #EpiPlex #CancerResearch #EMBO2025

RNA modifications such as m6A and inosine reveal a layer of gene regulation that traditional RNA sequencing cannot capture. In this new GEN Tutorial, Dr. Ananya Anmangandla shows how the EpiPlex™ platform and EpiScout™ analysis suite quantify both the genomic location and relative abundance of RNA modifications, even from ultra-low RNA inputs. Using a tumor–normal liver pair model for hepatocellular carcinoma (HCC), EpiPlex uncovered striking differences between global and gene-level methylation patterns. Global hypomethylation contrasted with localized hyper- or hypomethylation at specific transcripts, revealing how transcript-level resolution is key to RNA modification profiling. Notably, differentially methylated and differentially expressed genes only partially overlapped, highlighting how RNA modification profiling can uncover distinct regulatory mechanisms and new therapeutic targets.
